A formulation brief asks for sodium hyaluronate in a clear serum. The samples arrive in different forms: one is a high-molecular-weight powder, the other a low-molecular-weight solution. Where should the evaluation start?
The serum’s target viscosity is a useful place. A fluid toner, a serum with some body and a cream with its own thickening system each place different demands on the ingredient. The manufacturing team may also have a preference if preparing a powder requires extra processing time.
Sodium hyaluronate does more than contribute to hydration. In water, its molecular weight and concentration influence viscosity (Bothner and Wik, 1987). That can make a difference to the way a serum flows and spreads.
| Formulation Target | Starting Point for Evaluation | What to Check |
|---|---|---|
| Fluid toner or essence | A lower-viscosity grade or solution | Clarity, flow and skin feel |
| Serum with more body | A higher-molecular-weight grade | Viscosity, stringiness and spreadability |
| Cream with an established thickener | A grade selected for the desired use level and feel | Interaction with the existing rheology system |
| Multi-MW concept | A blend of molecular-weight grades | Blend composition, viscosity and finished-product performance |
Note: The table gives the lab somewhere to start. It cannot predict the feel of the whole formula. A low-MW grade may contribute little viscosity, for example, but glycerin and the thickening system can still leave the product feeling tacky.
Two materials described as “low molecular weight” may have different numerical ranges. Ask for those ranges and the test methods before comparing them. The same applies to a multi-MW blend: find out what the reported value represents.
Human testing also needs to be read in context. In a double-blind study, 36 older adults applied three lotions to separate areas of dry skin on their legs: a low-MW HA lotion, a high-MW HA lotion and a vehicle (Muhammad et al., 2024).
| Result after 4 weeks | Low-MW lotion | High-MW lotion | Vehicle |
|---|---|---|---|
| Skin capacitance, AU | 56.37 | 52.37 | 49.01 |
The low-MW lotion produced higher skin capacitance under the study conditions. There was no significant difference between groups in transepidermal water loss. The study supports the hydration performance of the tested lotions in this group. It does not establish that every low-MW raw material will outperform every high-MW grade. It also does not prove that a multi-MW blend is automatically more effective than a single grade.
The choice often comes down to manufacturing conditions.
| Question | Powder | Pre-made Solution |
|---|---|---|
| Preparation | Requires suitable dispersion and hydration | Already hydrated; still needs compatibility testing |
| Formula space | Adds relatively little water | Introduces water and possibly other ingredients |
| Storage & Transport | More concentrated | Larger volume for the same polymer content |
| Microbiological control | Check the grade’s limits and handling | Review microbial limits and preservation |
| Cost comparison | Check content as supplied | Calculate cost at the required solution addition level |
A pre-made solution can save the factory from preparing its own stock solution. Powder takes up less space in transport and adds little water to the formula, although it needs a suitable hydration process. Huatai supplies sodium hyaluronate in powder and solution forms. Its listed options include a standard 1% solution and multi-MW blends. Check the current documents for the particular grade before ordering a sample.
Take a formula that needs 0.05% sodium hyaluronate. Under the assumption that the powder contains 100% sodium hyaluronate as supplied, the powder addition would be 0.05%. The same target would require 5% of a 1% solution.
| Material | Assumed Content as Supplied | Addition Level | Sodium Hyaluronate Delivered |
|---|---|---|---|
| Powder | 100% | 0.05% | 0.05% |
| Solution | 1% | 5.00% | 0.05% |
This is a calculation example, not a product specification. Actual powder content may be lower. If the COA reports its assay on a dry basis, request the information needed to calculate the content of the powder as supplied. In the example, the 5% solution addition carries approximately 4.95% water and other components into the batch. Check those details before reducing the added water; include any preservative in the finished-product assessment.
A specification sets acceptance limits. A COA reports the results for a particular batch. Both are needed when assessing consistency.
Thermo Scientific lists one research-use sodium hyaluronate powder with a molecular weight of 1.5 to 2.2 million Da. Its specification sets the assay at at least 94% on a dry basis and loss on drying at no more than 10%. These are product limits, not results from a particular batch. The material is intended for research use, so the figures should not be treated as cosmetic-grade requirements.
When reviewing a cosmetic-grade sample, ask the supplier to clarify:
| Parameter | Question to Ask |
|---|---|
| Assay | Does the percentage refer to dried material or the powder as supplied? |
| Molecular weight | Which range does the grade cover, and which method was used? |
| Loss on drying | Is it reported for each batch? |
| Microbiology | What are the limits and the actual batch results? |
| Solution composition | What is present besides sodium hyaluronate and water? |
| pH and viscosity | What were the sample concentration and temperature during testing? |
Once the documents have narrowed the selection, put the sample into the intended base. Compare its appearance, pH, viscosity and skin feel with the formulation target. If using a solution, account for everything it adds to the preservative system.
